Hoveyda-Grubbs Catalyst 2nd Generation (CAS: 301224-40-8) is a phosphine free version of Grubbs 2nd Generation Catalyst with comparable reactivity, but initiates more readily at lower temperatures. Efficient for metathesis of electron-deficient substrates including fluorinated olefins. Demonstrated in multiple large-scale pharmaceutical applications.
Hoveyda-Grubbs Catalyst 2nd Generation can be used as a catalyst for olefin cross-metathesis with fluorinated olefins (CM), ring-closing metathesis (RCM), ring-opening metathesis (ROM), and a sequence of a metathesis reaction and subsequent dihydroxylation of the newly formed double bond.
In the first generation Hoveyda-Grubbs Catalyst, one of the phosphine ligands is replaced by an isopropyloxy group attached to the benzene ring. Its second generation has the other phosphine ligand replaced by NHC.
Grubbs catalyst has poor thermal stability and is prone to decomposition at higher temperatures. Amir H. Hoveyda et al. introduced large nucleophilic isopropoxychelate ligands into the molecule, which improved the thermal stability of the catalysts, which were called "Hoveyda-Grubbs catalysts". It is also divided into the first generation and the second generation, respectively, derived from the corresponding Grubbs catalyst, but one of the tricyclohexylphosphine groups in the original Grubbs catalyst is replaced by the isopropoxy group of the benzene ring. The disadvantage of these catalysts is that the catalytic activity is general, and it is difficult to separate the catalyst from the reaction system after the reaction.
The first generation Hoveyda-Grubbs catalyst is suitable for the loop closure complex decomposition of terminal olefin. The second generation Hoveyda-Grubbs catalyst is suitable for the ring closing, ring opening and cross decomposition reactions of electron-deficient alkenes.
Although the first and second generation Grubbs catalysts have high catalytic activity, some reactions still require a large amount, sometimes even 50mol% of the catalyst, and the catalyst can not be reused. In addition, the metal residues in the reaction products are difficult to remove in the post-treatment. Hoveyda catalysts are stable in air and water and can be prepared by chromatographic column separation.
