Fmoc-β-Alanine (Fmoc-β-Ala-OH) (CAS: 35737-10-1) is an Fmoc protected alanine derivative widely used in the synthesis of drugs and peptides. Reaction type: Fmoc solid-phase peptide synthesis.
Fmoc-amino acids, used in peptide synthesis, used as an organic synthesis intermediate, pharmaceutical intermediate, biochemical reagent or chemical reagent. Used as Amino acidnutrition.
Preparation method β-Alanine (β-Ala) (0.5g, 6.61mmol) was dissolved in 10% NaCO3 (12ml) under stirring in a 50ml flask. Gradually add Fmoc-Cl (1.45g, 5.61mmol) of dioxane (10ml) solution to the resulting solution that has been placed in an ice bath. The reaction mixture was stirred at room temperature for 4 hours, then water (80ml) was added. The effluent phase layer was separated from the reaction mixture and stripping with ether three times (75ml × 3). The stripping water layer was acidified with an aqueous solution of 2NHCl to a pH of 2, and then extracted with ethyl acetate three times (75ml × 3). The organic phase layer was recovered and concentrated to obtain 1.50g of crude product. The crude product was recrystallized in a mixed solvent (30ml) of ethyl acetate : hexane = 1:2, and filtered under reduced pressure to obtain a white solid, expressed as Fmoc-beta-Alanine.
