Glycine tert-Butyl Ester Hydrochloride (H-Gly-OtBu·HCl) (CAS: 27532-96-3) is not only an important chemical raw material, but also widely used in drug synthesis and has good development prospects.
Amino acids and derivatives, used in peptide synthesis, organic synthesis intermediates, pharmaceutical intermediates, biochemical reagents, chemical reagents.
Preparation 1400L t-Butyl Acetate, 105KG Glycine and 180KG Perchloric Acid were added dropwise at a uniform speed in a 2000L reaction tank. React at 0-10℃ for about 48-72 hours. TLC monitors the reaction during the reaction. After the glycine reaction is more than 90%, use 4 N sodium hydroxide aqueous solution to adjust the PH 6.5-7.5 to separate the organic layer. The water layer continues to be extracted with tert-Butyl Acetate. After the water product is basically extracted to the 95%, all product layers are combined, washed with brine to TLC purity to obtain 1500L tert-Butyl Glycine/tert-Butyl Ethyl Ester solution, concentrate and recover tert-Butyl Ethyl Ester to obtain 180KG of tert-Butyl Glycine, add 100L of Ethyl Ethyl Ester, pass hydrogen chloride to PH0.5, stir and cool the cold crystal, and centrifugally wash to obtain 182KG of tert-Butyl Glycine Hydrochloride.
