N-Hydroxysuccinimide is often used to assist carbodiimide-mediated peptide coupling by forming an active ester intermediate via condensation of the surface carboxyl group and NHS. The NHS-reactive ester intermediate is susceptible to nucleophilic attack by primary amines and results in the formation of stable amide bonds between the biomaterial surface and the N-terminus of the peptide.
N-Hydroxysuccinimide is a hydroxylated succinimide that is widely used in the activation of carboxylic acids to NHS-esters to promote coupling with nucleophiles. This approach is most commonly employed in activating the free acid forms of fluorophores for promoting conjugation of these fluorophores to amine residues of proteins. Use of NHS ester activation has also been described in the synthesis of N-acyl amino acids.
Used for the synthesis of amino acid protectant, semi-synthetic kanamycin and pharmaceutical intermediates.N-hydroxybutyldiimide and its derivatives are important raw materials for the synthesis of peptides, antibiotics, amino acids and proteins.N-hydroxybutyldiimide is used in the preparation of active esters, and the inhibition of the optically active effect of peptide coupling has a wide range of USES in medicine, and is also a basic chemical raw material.
N-Hydroxysuccinimide is used for improved amidations in the carbodiimide method. It is also used to activate a carboxyl group and reacts with amine to form amide. It is involved in the preparation of N-hydroxymaleimide-styrene copolymer.
N-Hydroxysuccinimide was prepared as follows: 100g (1.0mol) succinic anhydride and 70g (1.0mol) of hydroxylamine hydrochloride are heated together, the reaction system is kept under negative pressure to remove the generated volatiles, and is rapidly heated to above 125℃, and then slowly rises to 160℃ after 1H. The heating was stopped and when the temperature dropped to 125℃, the liquid reaction was poured into of diethyl ether and stirred vigorously. After the product was cured, the ether layer was decanted, the cured product was heated to boiling with 400ml of butanol, hot filtered, and the filtrate was rapidly cooled to 0℃. After 1H filtration, the crystals were successively washed with butanol and diethyl ether, and the obtained crude product was recrystallized from ethyl acetate to obtain 50g of finished product in 44% yield.
